Megazyme酶法分析检测原理

NADH((或者 NADPH)在340nm波长处具有强吸收峰,Megazyme酶法分析检测产品利用这一原理,选择消耗或者产生NADH((或者 NADPH)的化学反应,利用分光光度计检测NADH((或者NADPH)浓度的变化,从而达到检测待检物质浓度的目的。

Enzyme Substrates
Dye labelled polysaccharides are very useful chromogenic substrates for the measurement of endo-glycosyl hydrolase activity. Soluble chromogenic substrates are useful for the quantitative assay of enzyme solutions using the standard curve available for each product while insoluble chromogenic substrates are mostly employed qualitatively for the detection of specific enzyme activities in gels or plate screening experiments. Enzyme tablet tests contain the insoluble chromogenic substrates but in tablet form they can be used for quantitative assays with added convenience for the user. Colourimetric oligosaccharides are soluble, chemically defined, functionalised oligosaccharides that can be used for the specific assay of exo- or endo-glycosyl hydrolases and are suitable for use with automated analyzer systems.

This test kit is suitable for the measurement and analysis of Total Dietary Fiber as per Codex Alimentarius definition, updated to be more consistent with in vivo conditions in the human small intestine, i.e. a 4 h incubation time. Under these conditions more accurate measurement of resistant starch is obtained, including phosphate cross-liked starch (RS4). Use of higher enzyme concentrations ensures that resistant maltodextrins produced from non-resistant starch under the incubation conditions of the Integrated Total Dietary Fiber procedure (AOAC Methods 2009.01 and 2011.25 are no longer produced.
In this rapid, improved method, the incubation time with PAA + AMG is reduced to 4 h and the levels of both PAA and AMG are increased to ensure that resistant starch levels obtained with a set of control samples are consistent with ileostomy data. Under these conditions, the DF values obtained for most samples are the same as those obtained with AOAC Methods 2009.01 and 2011.25.
The dietary fiber fractions that are measured with this method are:
1. High Molecular Weight Dietary Fiber (HMWDF) including Insoluble Dietary Fiber (IDF) and Low Molecular Weight Soluble Dietary Fiber (SDFS; soluble dietary fiber which is soluble in the presence of 78% aqueous ethanol), or
2. Insoluble dietary fiber (IDF), Higher Molecular Weight Soluble Dietary Fiber (SDFP; water soluble dietary fiber that precipitates in the presence of 78% aqueous ethanol) and SDFS.
The enzymes used in this method are high purity and effectively devoid of contaminating enzymes active on other dietary fiber components such as β-glucan, pectin and arabinoxylan. They are supplied as freeze-dried powders; allowing the use of glycerol as an internal standard.

The Amylase SD Method is a highly sensitive colourimetric method for the determination of α-amylase in sprout damaged wheat grain (also known as pre-harvest sprouting or weather damaged wheat grain) and “late maturity α-amylase” wheat grain. Can also be used for measuring α-amylase in confectionery, soft drinks, brewing and fermentation, jams, sauces, conserves, ice cream and baby food.

For use with the Total Dietary Fiber Assay Kit. Contains barley β-glucan, high amylose maize starch, wheat starch, casein, pectin and larch galactan. Wheat arabinoxylan is available on request.

优点:

使用的酶为高纯度酶,酶之间没有干扰效应的,且具有标准化酶活力。Megazyme 淀粉葡糖甘酶完全没有纤维素酶的活性,但其他公司生产的酶常被此活性污染,那样会导致β-葡聚糖被溶解和含量经常被低估。所有的Megazyme酶都是液态即用型。

应用方法:

总膳食纤维是采用经过干燥和脱脂(如果脂肪含量>10%)的复样为样品。样品用热稳定的α-淀粉酶在100℃下处理以使淀粉成胶状,水解和解聚;用蛋白酶(使蛋白溶解和解聚)和淀粉葡糖甘酶(将淀粉碎片水解成葡萄糖(来自淀粉))在60℃下处理;然后用4倍体积的乙醇处理以沉淀可溶性纤维,去除解聚的蛋白和葡萄糖(来自淀粉)。然后残留物分别用78%乙醇,95%乙醇,丙酮洗涤,再干燥,称重。一份复样用于蛋白分析,另外一份在525℃下孵育以测定灰分,总TDF就是被过滤和干燥的残留物的重量减去蛋白和灰分的重量。

适用范围:

应用于谷粒,果蔬,谷物和水果型产品食品。
酶纯度和标准化:

α-淀粉酶(E-BLAAM)的酶活为3,000UmL(Ceralpha 法);蛋白酶的浓度为50mg/mL(~350 酪氨酸U/mL);淀粉葡糖甘酶单位为200U/mL(底物为对硝基苯基β-麦芽糖苷)或单位3300U/ mL(底物为可溶性淀粉),淀粉葡糖甘酶酶活为传统TDF分析使用的150%。
